GLP-2TZ is a synthetic 39-amino acid linear peptide agonist of both the glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP) receptors, featuring a C20 fatty diacid moiety conjugated to an amino acid residue at position 20 via a linker to extend its half-life to approximately 5 days, enabling once-weekly subcutaneous administration. Derived from modifications to the native GIP sequence with GLP-1-like pharmacological activity, it exhibits biased signaling, particularly at the GLP-1 receptor, promoting glucose-dependent insulin secretion, suppression of glucagon release, enhanced insulin sensitivity, and delayed gastric emptying, while its GIP agonism augments β-cell function and contributes to superior weight loss effects compared to GLP-1 monotherapy.  GLP-2tz demonstrates robust reductions in HbA1c (up to 2.3%) and body weight  with additional benefits in cardiovascular risk factors and potential renoprotective effects.
